Let's suppose I have an object made of a single face, say a square laid down on the grid. Can I manually select an edge, then programatically, using the API, insert three vertices somewhere between the two points that define the edge's extremities? That might change the face's shape, of course.

Yes. You'd have to create a replacement for the whole face with the new points and then delete the original face.
